چکیده

Perfect timing of germination is required to encounter optimal conditions for plant survival and is the result of a complex interaction between molecular processes, seed characteristics, and environmental cues. To detangle these processes, we made use of natural genetic variation present in an Arabidopsis (Arabidopsis thaliana) Bayreuth × Shahdara recombinant inbred line population. For a detailed analysis of the germination response, we characterized rate, uniformity, and maximum germination and discuss the added value of such precise measurements. The effects of after-ripening, stratification, and controlled deterioration as well as the effects of salt, mannitol, heat, cold, and abscisic acid (ABA) with and without cold stratification were analyzed for these germination characteristics. Seed morphology (size and length) of both dry and imbibed seeds was quantified by using image analysis. For the overwhelming amount of data produced in this study, we developed new approaches to perform and visualize high-throughput quantitative trait locus (QTL) analysis. We show correlation of trait data, (shared) QTL positions, and epistatic interactions. The detection of similar loci for different stresses indicates that, often, the molecular processes regulating environmental responses converge into similar pathways. Seven major QTL hotspots were confirmed using a heterogeneous inbred family approach. QTLs colocating with previously reported QTLs and well-characterized mutants are discussed. A new connection between dormancy, ABA, and a cripple mucilage formation due to a naturally occurring mutation in the MUCILAGE-MODIFIED2 gene is proposed, and this is an interesting lead for further research on the regulatory role of ABA in mucilage production and its multiple effects on germination parameters.

LAGA: A Software for Landscape Allocation using Genetic Algorithm

In this paper, Landscape Allocation using Genetic Algorithm (LAGA), a spatial multi-objective land use optimization software is introduced. The software helps in searching for optimal land use when multiple objectives such as suitability, area, cohesion and edge density indices are simultaneously involved. LAGA is a flexible and easy to use genetic algorithm-based software for optimizing the sp...

Improved salt tolerance in canola (Brasica napus) plants by overexpression of Arabidopsis Na+/H+ antiporter gene AtNHX1

A significant portion of the world’s cultivated land is affected by salinity that reduces crop productivity in these areas. Breeding for salt tolerance is one of the important strategies to overcome this problem. Recently, genetic engineering is becoming a promising approach to improving salt tolerance. In order to improve the yield performance of canola in saline soils, we transformed canola w...

Factors affecting delivery of DREB1A gene in maize B73 split-seeds via biolistic system

Immature embryos as a choice tissue for genetic transformation of maize have a few limitations, such as genotype dependence, time-consuming and requiring a well-equipped greenhouse for access, at any time. In the present study, the split-seed explants were used for genetic transformation of maize, B73 line. The transformation of maize split-seed explants from the inbred line B73, for resistance...

Demographic and Genetic Patterns of Variation among Populations of Arabidopsis thaliana from Contrasting Native Environments

BACKGROUND Understanding the relationship between environment and genetics requires the integration of knowledge on the demographic behavior of natural populations. However, the demographic performance and genetic composition of Arabidopsis thaliana populations in the species' native environments remain largely uncharacterized. This information, in combination with the advances on the study of ...

Differential Expression of Arabidopsis thaliana Acid Phosphatases in Response to Abiotic Stresses

The objective of this research is to identify Arabidopsis thaliana genes encoding acid phosphatases induced by phosphate starvation. Multiple alignments of eukaryotic acid phosphatase amino acid sequences led to the classification of these proteins into four groups including purple acid phosphatases (PAPs). Specific primers were degenerated and designed based on conserved sequences of PAPs isol...
